    SOME FACTORS INFLUENCING THE FREE FLUORIDE CONTENT IN BLACK TEA INFUSIONS

    Fluoride is an essential micro-nutrient owing to its role in the prevention of dental caries. Chronic exposure to high levels of the ion (F-) results in both dental and skeletal fluorosis. A study was carried out to determine free fluoride content in tea ( Camellia sinensis ) infusions of different grades of black Cut, Tear and Curl (CTC) tea. Tea infusions, 1% w/v of 112 teas of different grades and origins, were prepared using boiling distilled water and their free fluoride contents quantified using a Fluoride Ion Selective Electrode (FISE) method. The free fluoride content of the teas was 0.37\ub10.04 \ub5g F- ml-1. Infusions of tea grades BP1 and BMF gave the highest (0.40\ub10.22 \ub5g F- ml-1) and lowest (0.32\ub10.10 \ub5g F- ml-1) free fluoride contents, respectively. There being no national and international Maxima Residue Limit (MRL) for fluoride in tea, all the teas analysed may be regarded to be safe for consumption with regard to their free fluoride contents based on the 1.5 \ub5g F- ml-1, World Health Organisation\u2019s (WHO) guideline for fluoride in drinking water. Further, the effects of tea grades and area of production, on the fluoride content in tea infusions were significant (P<0.05), hence the need to enact safety guidelines and monitor continuously the fluoride levels for all types of tea products to prevent excessive intake.Le Fluor est un micro-\ue9l\ue9ment essentiel lorsque consomm\ue9 en dessous de certains seuils eu \ue9gard \ue0 son r\uf4le dans la pr\ue9vention des caries dentaires.     Crystal structure and proton conductivity of BaSn0.6Sc0.4O3-delta: insights from neutron powder diffraction and solid-state NMR spectroscopy

    The solid-state synthesis and structural characterisation of perovskite BaSn(1–x)Sc(x)O(3–δ) (x = 0.0, 0.1, 0.2, 0.3, 0.4) and its corresponding hydrated ceramics are reported. Powder and neutron X-ray diffractions reveal the presence of cubic perovskites (space group Pm3m) with an increasing cell parameter as a function of scandium concentration along with some indication of phase segregation. (119)Sn and (45)Sc solid-state NMR spectroscopy data highlight the existence of oxygen vacancies in the dry materials, and their filling upon hydrothermal treatment with D(2)O. It also indicates that the Sn(4+) and Sc(3+) local distribution at the B-site of the perovskite is inhomogeneous and suggests that the oxygen vacancies are located in the scandium dopant coordination shell at low concentrations (x ≤ 0.2) and in the tin coordination shell at high concentrations (x ≥ 0.3). (17)O NMR spectra on (17)O enriched BaSn(1–x)Sc(x)O(3–δ) materials show the existence of Sn–O–Sn, Sn–O–Sc and Sc–O–Sc bridging oxygen environments. A further room temperature neutron powder diffraction study on deuterated BaSn(0.6)Sc(0.4)O(3–δ) refines the deuteron position at the 24k crystallographic site (x, y, 0) with x = 0.579(3) and y = 0.217(3) which leads to an O–D bond distance of 0.96(1) Å and suggests tilting of the proton towards the next nearest oxygen. Proton conduction was found to dominate in wet argon below 700 °C with total conductivity values in the range 1.8 × 10(–4) to 1.1 × 10(–3) S cm(–1) between 300 and 600 °C. Electron holes govern the conduction process in dry oxidizing conditions, whilst in wet oxygen they compete with protonic defects leading to a wide mixed conduction region in the 200 to 600 °C temperature region, and a suppression of the conductivity at higher temperature

    The Endogenous Th17 Response in NO<inf>2</inf>-Promoted Allergic Airway Disease Is Dispensable for Airway Hyperresponsiveness and Distinct from Th17 Adoptive Transfer

    Severe, glucocorticoid-resistant asthma comprises 5-7% of patients with asthma. IL-17 is a biomarker of severe asthma, and the adoptive transfer of Th17 cells in mice is sufficient to induce glucocorticoid-resistant allergic airway disease. Nitrogen dioxide (NO2) is an environmental toxin that correlates with asthma severity, exacerbation, and risk of adverse outcomes. Mice that are allergically sensitized to the antigen ovalbumin by exposure to NO2 exhibit a mixed Th2/Th17 adaptive immune response and eosinophil and neutrophil recruitment to the airway following antigen challenge, a phenotype reminiscent of severe clinical asthma. Because IL-1 receptor (IL-1R) signaling is critical in the generation of the Th17 response in vivo, we hypothesized that the IL-1R/Th17 axis contributes to pulmonary inflammation and airway hyperresponsiveness (AHR) in NO2-promoted allergic airway disease and manifests in glucocorticoid-resistant cytokine production. IL-17A neutralization at the time of antigen challenge or genetic deficiency in IL-1R resulted in decreased neutrophil recruitment to the airway following antigen challenge but did not protect against the development of AHR. Instead, IL-1R-/- mice developed exacerbated AHR compared to WT mice. Lung cells from NO2-allergically inflamed mice that were treated in vitro with dexamethasone (Dex) during antigen restimulation exhibited reduced Th17 cytokine production, whereas Th17 cytokine production by lung cells from recipient mice of in vitro Th17-polarized OTII T-cells was resistant to Dex. These results demonstrate that the IL-1R/Th17 axis does not contribute to AHR development in NO2-promoted allergic airway disease, that Th17 adoptive transfer does not necessarily reflect an endogenously-generated Th17 response, and that functions of Th17 responses are contingent on the experimental conditions in which they are generated. © 2013 Martin et al

    A longitudinal cohort study of malaria exposure and changing serostatus in a malaria endemic area of rural Tanzania.

    BACKGROUND: Measurements of anti-malarial antibodies are increasingly used as a proxy of transmission intensity. Most serological surveys are based on the use of cross-sectional data that, when age-stratified, approximates historical patterns of transmission within a population. Comparatively few studies leverage longitudinal data to explicitly relate individual infection events with subsequent antibody responses. METHODS: The occurrence of seroconversion and seroreversion events for two Plasmodium falciparum asexual stage antigens (MSP-1 and AMA-1) was examined using three annual measurements of 691 individuals from a cohort of individuals in a malaria-endemic area of rural east-central Tanzania. Mixed-effect logistic regression models were employed to determine factors associated with changes in serostatus over time. RESULTS: While the expected population-level relationship between seroprevalence and disease incidence was observed, on an individual level the relationship between individual infections and the antibody response was complex. MSP-1 antibody responses were more dynamic in response to the occurrence and resolution of infection events than AMA-1, while the latter was more correlated with consecutive infections. The MSP-1 antibody response to an observed infection seemed to decay faster over time than the corresponding AMA-1 response. Surprisingly, there was no evidence of an age effect on the occurrence of a conversion or reversion event. CONCLUSIONS: While the population-level results concur with previously published sero-epidemiological surveys, the individual-level results highlight the more complex relationship between detected infections and antibody dynamics than can be analysed using cross-sectional data. The longitudinal analysis of serological data may provide a powerful tool for teasing apart the complex relationship between infection events and the corresponding immune response, thereby improving the ability to rapidly assess the success or failure of malaria control programmes

    Toward a Surrogate Marker of Malaria Exposure: Modeling Longitudinal Antibody Measurements under Outbreak Conditions

    Background: Biomarkers of exposure to Plasmodium falciparum would be a useful tool for the assessment of malaria burden and analysis of intervention and epidemiological studies. Antibodies to pre-erythrocytic antigens represent potential surrogates of exposure. Methods and Findings: In an outbreak cohort of U.S. Marines deployed to Liberia, we modeled pre- and post-deployment IgG against P. falciparum sporozoites by immunofluorescence antibody test, and both IgG and IgM against the P. falciparum circumsporozoite protein by enzyme-linked immunosorbant assay. Modeling seroconversion thresholds by a fixed ratio, linear regression or nonlinear regression produced sensitivity for identification of exposed U.S. Marines between 58-70% and specificities between 87-97%, compared with malaria-naïve U.S. volunteers. Exposure was predicted in 30-45% of the cohort. Conclusion: Each of the three models tested has merits in different studies, but further development and validation in endemic populations is required. Overall, these models provide support for an antibody-based surrogate marker of exposure to malaria

    High-Utilisation Nanoplatinum Catalyst (Pt@cPIM) Obtained via Vacuum Carbonisation in a Molecularly Rigid Polymer of Intrinsic Microporosity

    Polymers of intrinsic microporosity (PIM or here PIM-EA-TB) offer a highly rigid host environment into which hexachloroplatinate(IV) anions are readily adsorbed and vacuum carbonised (at 500 °C) to form active embedded platinum nanoparticles. This process is characterised by electron and optical microscopy, atomic force microscopy (AFM), X-ray photoelectron spectroscopy (XPS) and electrochemical methods, which reveal that the PIM microporosity facilitates the assembly of nanoparticles of typically 1.0 to 2.5-nm diameter. It is demonstrated that the resulting carbonised “Pt@cPIM” from drop-cast films of ca. 550-nm average thickness, when prepared on tin-doped indium oxide (ITO), contain not only fully encapsulated but also fully active platinum nanoparticles in an electrically conducting hetero-carbon host. Alternatively, for thinner films (50–250 nm) prepared by spin coating, the particles become more exposed due to additional loss of the carbon host. In contrast to catalyst materials prepared by vacuum-thermolysed hexachloroplatinate(IV) precursor, the platinum nanoparticles within Pt@cPIM retain high surface area, electrochemical activity and high catalyst efficiency due to the molecular rigidity of the host. Data are presented for oxygen reduction, methanol oxidation and glucose oxidation, and in all cases, the high catalyst surface area is linked to excellent catalyst utilisation. Robust transparent platinum-coated electrodes are obtained with reactivity equivalent to bare platinum but with only 1 μg Pt cm−2 (i.e. ~100% active Pt nanoparticle surface is maintained in the carbonised microporous host).     Cord blood IgG and the risk of severe Plasmodium falciparum malaria in the first year of life

    Young infants are less susceptible to severe episodes of malaria but the targets and mechanisms of protection are not clear. Cord blood antibodies may play an important role in mediating protection but many studies have examined their association with the outcome of infection or non-severe malaria. Here, we investigated whether cord blood IgG to Plasmodium falciparum merozoite antigens and antibody-mediated effector functions were associated with reduced odds of developing severe malaria at different time points during the first year of life. We conducted a case-control study of well-defined severe falciparum malaria nested within a longitudinal birth cohort of Kenyan children. We measured cord blood total IgG levels against five recombinant merozoite antigens and antibody function in the growth inhibition activity and neutrophil antibody-dependent respiratory burst assays. We also assessed the decay of maternal antibodies during the first 6months of life. The mean antibody half-life range was 2.51months (95% confidence interval (CI): 2.19-2.92) to 4.91months (95% CI: 4.47-6.07). The rate of decline of maternal antibodies was inversely proportional to the starting concentration. The functional assay of antibody-dependent respiratory burst activity predicted significantly reduced odds of developing severe malaria during the first 6months of life (Odds ratio (OR) 0.07, 95% CI: 0.007-0.74, P=0.007). Identification of the targets of antibodies mediating antibody-dependent respiratory burst activity could contribute to the development of malaria vaccines that protect against severe episodes of malaria in early infancy

    Sero-epidemiological evaluation of changes in Plasmodium falciparum and Plasmodium vivax transmission patterns over the rainy season in Cambodia

    <p>Abstract</p> <p>Background</p> <p>In Cambodia, malaria transmission is low and most cases occur in forested areas. Sero-epidemiological techniques can be used to identify both areas of ongoing transmission and high-risk groups to be targeted by control interventions. This study utilizes repeated cross-sectional data to assess the risk of being malaria sero-positive at two consecutive time points during the rainy season and investigates who is most likely to sero-convert over the transmission season.</p> <p>Methods</p> <p>In 2005, two cross-sectional surveys, one in the middle and the other at the end of the malaria transmission season, were carried out in two ecologically distinct regions in Cambodia. Parasitological and serological data were collected in four districts. Antibodies to <it>Plasmodium falciparum </it>Glutamate Rich Protein (GLURP) and <it>Plasmodium vivax </it>Merozoite Surface Protein-1<sub>19 </sub>(MSP-1<sub>19</sub>) were detected using Enzyme Linked Immunosorbent Assay (ELISA). The force of infection was estimated using a simple catalytic model fitted using maximum likelihood methods. Risks for sero-converting during the rainy season were analysed using the Classification and Regression Tree (CART) method.</p> <p>Results</p> <p>A total of 804 individuals participating in both surveys were analysed. The overall parasite prevalence was low (4.6% and 2.0% for <it>P. falciparum </it>and 7.9% and 6.0% for <it>P. vivax </it>in August and November respectively). <it>P. falciparum </it>force of infection was higher in the eastern region and increased between August and November, whilst <it>P. vivax </it>force of infection was higher in the western region and remained similar in both surveys. In the western region, malaria transmission changed very little across the season (for both species). CART analysis for <it>P. falciparum </it>in the east highlighted age, ethnicity, village of residence and forest work as important predictors for malaria exposure during the rainy season. Adults were more likely to increase their antibody responses to <it>P. falciparum </it>during the transmission season than children, whilst members of the Charay ethnic group demonstrated the largest increases.</p> <p>Discussion</p> <p>In areas of low transmission intensity, such as in Cambodia, the analysis of longitudinal serological data enables a sensitive evaluation of transmission dynamics. Consecutive serological surveys allow an insight into spatio-temporal patterns of malaria transmission. The use of CART enabled multiple interactions to be accounted for simultaneously and permitted risk factors for exposure to be clearly identified.</p
